Abstract of the DisclosureIt is known to determine the concentration of glucose in abiological sample using a number of different spectrophotometric indicators.However the known indicators have a number of drawbacks in that sensitivespectrometers are required for the test, and usually the sample must have theprotein removed from it. The present invention seeks to overcome thesedrawbacks by providing a process for the enzymatic spectrophotometricdetermination of glucose in a sample comprising the oxidation of glucose inthe presence of an enzyme reagent therefore and the reaction of the resultinghydrogen peroxide with an indicator system comprising at least (a) compoundselected from (i) phenol which is unsubstituted or substituted by one or moreof hydrogen, halogen, NH2, SO?, C1 - C3 alkyl, cyclohexyl, methoxy, carboxylicacid, lower alkyl carboxylic ester, aldehyde,phenyl which may be unsubstitutedor further substituted by one or more of hydroxy, carbonyl or carboxylic acidor a radical ; (ii) naphthalene which is unsubstituted or substitutedby one or more of hydroxyl, methyl or SO?; or (iii) quinoline which isunsubstituted or substituted by one or more of hydroxyl; CH3; SO?; or(iv) barbituric acid which may not be substituted in its 5 position; 5pyrazolone; diketohydrindene; or 2,4-diamino-6-hydroxypyrimidine; and (b)a compound 4-aminoantipyrine. The indicator system furtherincludes ferrocyanide.
