Process for insulation ting and purifying a fraction capable of inhibiting mutagenicity of a mutagenic material from juice

PURPOSE:To obtain a simple substance having a high inhibitory activity of mutagen, by separating and purifying a mutagen inhibitor present in a burdock juice through a specific stage. CONSTITUTION:A burdock juice is centrifuged or ultracentrifuged to give a supernatant liquid, which is mixed with phosphoric acid buffer solution. The mixture is then salted with a water-soluble inorganic alkali metal salt, e.g. ammonium sulfate,. The salted mixture is centrifuged to give a prepcipitate, which is dissolved in a phosphoric acid buffer solution and dialyzed with water or phosphoric acid buffer solution, and the resultant dialyzate is ultrafiltered. The concentrate thus obtained is taken out or powdered by freeze-drying to afford an inhibitor of a mutagen. The inhibitor is a substance having the protein structure which is inactivated by manganese ion and stable to heat, has a strong anionic group and a maximum absorption at a wave length of 280nm. The resultant inhibitor is capable of inhibiting the mutagen, e.g. AF-2 or Trp-P-1 in the scorched part of roasted meat, with a high activity due to the high purity of the isolated substance.
