PURPOSE:To analyze quantitatively singularly combined protein in high accuracy, by bringing an excessive quantity of physiologically very small quantity active substance such as hormone in blood serum is brought into contact with protein singularly combining with said substance and utilizing a competitive reaction of labelled substance of said active substance, protein an unlabelled substance. CONSTITUTION:In order to analyze quantitatively a protein P in a sample to be inspected (blood serum etc.) containing protein (the protein P) singularly combining with physiologically very small quantity active substance (substance A) such as hormone in blood, metals etc., a complex A-P is formed by adding the substance A to the sample exceeding the combination capacity with the protein P. Next, the compex A-P is separated from the excess substance A and is brought into contact with the substance A labelled by a fixed quantity of a radioactive element or enzyme and the substitution is carried out by performing a competitive reaction of labelled substance A of the complex A-P and unlabelled substance A of the complex A-P. Next, after labelled complex A-P is separated from uncombined labelled substance A, radioactivity of separated labelled complex A-P is measured and the protein P is determined accurately. The complex A-P is separated more easily by adding a singular antibody for the protein P together with the protein P and combining said antibody with the complex A-P.
