Antihypertensive extract from olive leaves - contg. oleuropeine, using methanol as extracting solvent

An extract rich in oleuropeine is obtd. from dried olive leaves (Olea europea) by the following process. The leaves are refluxed in methanol, the solid eliminated, and the methanol concentrated. It is then diluted with water and washed with methylene chloride. After elimination of the organic phase the aq. soln. is extracted with ethyl acetate. The organic solution is dried and evaporated. 1-50 litres of methanol per kg dried leaves are used, pref. 5-20 litres. Extn. may be a single operation or a series of successive operations. The solid is sepd. by filtration, decantation or centrifuging. Concn. is effected at 30-50 deg. C, and diluted with 5-10 volumes of water. It is washed with 0.1-1.5 volumes of methylene chloride, then extracted with 0.5-5 volumes ethyl acetate, pref. 1-2 volumes. The aq. phase is removed by decantation.
