摘要

PURPOSE:To produce a large amount of amino acid, by using a plasmid vector connected with a gene coding the phosphoenolpyruvate carboxylase (PEPC) playing an important role in the supply of aspartic acid, introducing the plasmid vector into a coryneform, and culturing the coryneform. CONSTITUTION:The chromosome gene of a coryneform represented by Brevibacterium lactofermentum ATCC13869, etc. is extracted, and incised with a proper restriction enzyme. The cleaved gene is connected with a plasmid vector (e.g. pAM330, pAJ655, etc.) capable of proliferating in a coryneform, and a PEPC- deficient mutant of a coryneform is transformed with the resultant recombinant DNA. In the case of preparing a lysine-producing strain, a homoserine-requiring strain is used as the recombinant DNA-accepting strain, and in the case of producing arginine, 2-thiazolealanine-resistant strain is used as the accepting strain.
