PURPOSE:To perform fractional quantitative determination of amylazes with high accuracy by adding an amylase activity inhibitor to a sample, measuring the activity occuring in the difference in the inhibiting effect to the S type and P type amylase in the sample by using a specific enzyme electrode and determining quantiatively the amylase from said activity and the activity value when the inhibitor is not added. CONSTITUTION:The alpha-amylase activator refined from wheat is added to a sample of a blood, etc. contg. the P type amylase originating in the pancreas and the S-type amylase originating in the salivery gland and the residual amylase activity value R is measured. The total activity value T when no inhibitor is added to the same sample is measured. The respective measurement is accomplished by injecting the inhibitor into a reaction chamber formed by putting a buffer soln. 19 into a reaction cell 9 provided with an enzyme electrode 8 coated with an immobilized glucose oxidase film 6 on a hydrogen perioxide electrode, injecting a liquid mixture composed of glucoamylase and modified starch such as sodium starch glycolate therein after specified time and detecting the amt. R of the glucose formed after the injection from a change in the current of the electrode 8. The amt. T of the glucose formed when the inhibitor is not added to the same sample is measured and both amylases are fractionally and quantitatively determined from the calibration curve of the R/T and P type/S type ratio obtd. preliminarily with good accuracy.
