Total or conjugated bilirubin determn. - using bilirubin oxidase or laccase, opt. in presence of surfactant, aromatic-carboxylic acid, sulpha drug or protease
In the quantitative determn. of conjugated bilirubin in biological fluids, (A) aq. bilirubin solns. with different bilirubin concns. are reacted with bilirubin oxidase originating from a microorganism of the genus Myrothecium to give a calibration curve showing the relationship between the bilirubin concn. and the absorption of the reaction mixt. (B) a biological fluid with unknown bilirubin concn. is reacted with Myrothicium bilirubin oxidase and the absorption of the resulting reaction mixt. is measured; and (C) the bilirubin concn. in the biological fluid is calculated by comparison of the measured absorption with the calibration curve. In an alternative process, the above steps are effected but instead of the Myrothecium bilirubinase there is used any bilirubin oxidase or laccase enzyme in presence of a surfactant an aromatic carboxylic acid, a sulpho drug or a protase. Analysis of conjugated and unconjugated bilirubin is used in the clinical diagnosis of liver cell lesions, hepatitic jaundice, posthepatitic jaundice, liver dysfunction, etc.
