Detection of beta-haemolytic streptococcus antigens in a clinical specimen on a swab without centrifugation comprises (a) contact of the swab with aq. NaNO2 and glacial acetic acid to form HNO2 so that the antigen is made more accessible; (b) adjustment to pH 4.5-7.2 with buffer; (c) removal of part of the buffered soln.; (d) mixing of the removed soln. with minute latex particles coated with beta-haemolytic streptococcal antibodies; and (e) observation of the mixt. for formation of an agglutination lattice, which indicates the presence of the antigens.
