PURPOSE:To facilitate the making of an antigen for ELISA (enzyme labelling immunoassay), by refining an antigen utilizing a mouse IgA monoclone antibody for recognizing about 46Kd (K dalton) by mol. of M.hp. (mycoplasma hyopneumonia) antigen to measure an antibody against the M.hp. CONSTITUTION:First, a hybridoma is obtained to a produce a monoclone antibody against about 46Kd by mol. of M.hp. antigen. An abdominal dropsy obtained by vaccinating a mouse with the hybridoma thus obtained is diluted to a moderate density to be adsorbed on a plate for ELISA and after it is washed, a M.hp. antigen made soluble by Tween20 is added thereto to be sensitized at a room temperature for 2hr. The antigen left from the reaction with the monoclone antibody is removed by a washing and a serum to be inspected is added to the product. After a washing, an anti-pig IgG peroxidase-labelled serum is added thereto to be sensitized for 2hr and then, after a washing, an enzyme substrate is added to the mixture to react. After the stopping of the reaction, the absorbance (OD492nm) is measured.
