The subject of this invention is a diagnostic method for observing the deficiency or surplus of cells belonging to a specific T-cell population OKT6+ in humans. Cells belonging to this T-cell population are seen in amounts below normal in primary cirrhosis of the liver, multiple sclerosis and hyper-IgE and in amounts above normal in acute organ transplant and a complete lack in myasthenia gravis, acute mononucleosis infection, all phases of Hodgkin's disease and psoriasis. The method uses a new IgG-class monoclonal antibody whicha) reacts with about 70% of the normal human thymocytes, but not with normal human peripheral T-cells, B-cells, null cells or bone marrow cells,b) reacts with regular human thymocytes with specific means displayed in the figure,c) reacts with peripheral T-cell thymocytes with specific means displayed,d) reacts with different phased ALL-T-cells with specific means displayed,e) reacts with cell lines CEM and MOLT-4, but not with cell line HSB-2, andf) defines T-cell population OKT6+The antibody is produced by a hybridoma, prepared by integrating mouse myeloma cell line cells with spleen cells of a mouse immunized with human thymocytes.
