摘要

PURPOSE:To obtain the titled DNA, having excellent heat resistance and capable of advantageously producing malic acid, by cultivating a specific transformant microorganism. CONSTITUTION:A chromosomic DNA obtained by purifying Bacillus.sterothermo philus is cleaved with a restriction enzyme and then linked to a vector DNA (e.g. PUC12) cleaved with the restriction enzyme to afford a recombinant DNA, which is subsequently introduced into a host microorganism [e.g. Escherichia coli (E. coli) JM103 strain] to provide a transformant. The obtained transformant is then screened to provide a transformant microorganism, capable of coding thermostable malic enzyme and derived from the Bacillus.sterothermophilus. The resultant microorganism is then cultivated to collect the aimed malic enzyme accumulated in the microbial cells.
