METHOD OF HEIGHTENING SENSITIVITY OF ASSAY FOR TARGET LIGAND

PURPOSE: To detect the binding site on a ligand by treating the ligand to remove peripheral substances inhibiting the binding site thereon thereby treating the ligand existing in a humor sample, e.g. whole blood or urine, directly. CONSTITUTION: Ligand is treated to remove peripheral substances inhibiting the binding site thereon thus exposing the binding site. Immunological conjugation with a target ligand existing in a fluid sampled from a mammal in unpurified state is enhanced. More specifically, inhibitory substances are removed through digestion with enzyme or exposure to a mild acid and an additional binding site is exposed. Neuraminidase is an enzyme preferable for treating ligand. The treated ligand is used in immunoassay and an anti-ligand binding is detected. A labeled anti-ligand touches a treated ligand and a bond anti-ligand is detected.
