PURPOSE:To obtain the plate for ELISA which is immobilized with inactivated Newcastle disease virus (NDV) antigens without degrading the conjugation power to antibodies by coating plural pieces of the wells formed on the plate with an NDV particle soln. solubilized by an octylglucoxide soln. treatment. CONSTITUTION:The allantoic fluid collected from an embryonated egg inoculated with the NDV is centrifugally separated to obtain a supernatant and after the supernatant is subjected to sucrose gradient centrifugation, the supernatant is suspended again in a PBS (phosphoric acid buffer soln.) and is subjected to ultracentrifugation to obtain the refined NDV particles. The NDV particles are solubilized at a room temp. when the PBS soln. of the octylglucoxide is added to the soln. prepd. by suspending the NDV particles in the PBS. Many antigens 2 exist in the exosporium 1 of the NDV and the exosporium 1 is cut in the X-marked parts when solubilized and, therefore, the many antigens 2 participate in the conjugation to the antibodies at the time when the antigens are immobilized to the plate. The degradation of the conjugation power of the NDV with the antigens is thus obviated. The ELISA (enzyme immoassay) is thus safely and easily executed.
