PURPOSE:To obtain a physiologically active polysaccharide RON having specified physical and chemical properties, and antitumor, immunity regulating, and infection protecting activities by adding a polar solvent to an extract obtained by immersing rice bran in a sucrose solution of a specified concentration and treating the rice bran with hot water and separating and purifying the formed precipitate. CONSTITUTION:A polar organic solvent is added to an extract obtained by immersing rice bran in a sucrose solution of a concentration of 5-30%, keeping the solution for 1-7 days and extracting the rice bran suspension with hot water. The formed precipitate is collected and optionally purified to obtain a physiologically active polysaccharide RON having the following physical and chemical properties: (A) properties: a white non-crystalline powder which is tasteless and odorless, (B) solubility: soluble in water to form a milky viscous solution in a high concentration, soluble in formamide and insoluble in alcohol, acetone, etc., (C) pH of an aqueous solution: neutral to slightly acidic, (D) constituent saccharide: entirely composed of glucose, (E) elementary analysis: 44.0-45.0% C and 6.1-6.3% H, (F) structure: alpha-glucan having a main chain of 1,6-bonds and having a small amount of a 1,3,6-branched structure, (G) MW=20,000,000, (H) color reaction: positive to anthrone-nitric acid reaction, etc., and negative to biuret reaction, etc.
