CYCLOHEXIMIDE SENSITIVITY FACTOR OF YEAST, NEW DNA TO CODE THE SAME FACTOR AND PRODUCTION THEREOF

PURPOSE:To obtain a large amount of protein synthesis factor by culturing a transformant bacterium prepared by transduction of specific plasmid in a medium. CONSTITUTION:Cells prepared by culturing Saccharomyces carlsbergenis IFO 1,265 strain are extracted to give mRNA, which is combined with commentary DNA to give plasmid (A). Then a transformant of Escherichia coli prepared by transduction of the component A is extracted to give plasmid DNA (B) containing a base sequence to code cycloheximide sensitivity factor(CHSF) of yeast shown by the formula. Then the component B is transformed with Bacillus subtilis, etc., to give a transformant strain, which is cultured to give a culture mixture (C). Then the component C is centrifuged to give cells, which are ground, extracted and purified to yield protein of cycloheximide sensitivity factor.
