PURPOSE:To immobilize an antibody protein retaining high antigen-antibody reaction activity, by bringing the antibody protein into contact with a specific lipid monomolecular film developed on the surface of an aq. phase to form an antibody protein-monomolecular film complex film and then transferring the formed complex film to a solid substrate. CONSTITUTION:A 14-22C long chain fatty acid (e.g., stearic acid) and/or an ester thereof are dissolved in an organic solvent such as chloroform and then developed on the surface of distilled water or an aq. soln. contg. polyvalent metal salt (e.g. barium chloride) and having ordinarily 6.5-7.5pH to form a monomolecular film of the polyvalent metal salt and/or ester of the 14-22C long chain fatty acid. The formed monomolecular film is compressed into a surface pressure of 1-30mN/m. Subsequently, an antibody protein (e.g., immunoglobulin G) is injected into the aq. phase under the monomolecular film and allowed to stand for ordinarily 30min-1hr to form an antibody protein- monomolecular film complex film. The formed complex film is compressed into a surface pressure of 30-50mN/m and then transferred to a hydrophobized solid substrate such as quartz.
