METHOD FOR MECHANICAL ACTION DETERMINATION OF INHIBITOR ON REPRODUCTION VIRUS HERPES SIMPLEX IN CELL CROUP

The method is applied in medicine. It is used to determine exactly and precisely the mechanism of the inhibitor anti-herpes influence at virus protein level. The method involves the contamination of cell culture with Virus Herpes Simplex, treatment with an inhibitor, disintegration of the cells and two consecutive centrifugations (low and high-revolution) for virus concentration. Electrophoresis follows of the virus proteins in 10 per cent polyacrylamide gels in proportion methylene -bisacrylamide 100:1, electro-transfer of the proteins on a nylon membrane, blocking with 10 per cent skimmed dry milk, consecutive treatment with a specific serum, goat antihuman biotinilized IgG antibodies, avidinperoxidase and subsequent washing, substrate buffer solution addition and rinsing again.IMAGE
