Process for demonstrating the affinity of fibrinogen and its derivatives for the filamentous forms of yeasts, use in the field of the determination of pathogenic yeasts especially in haemoculture
The present invention relates to a process for demonstrating the possible affinity of a substance (F) chosen from the set consisting of Fg, Fn, FDP and mixtures thereof, towards the filamentous forms of yeast strains, the said process being characterised in that it comprises the steps consisting in (1) culturing (A) a sample containing test yeasts essentially in the form of blastospores, which sample is capable of containing at least one substance which may interfere with the mechanism of adhesion of the said substance (F) onto the said yeasts and is suitably diluted in order to remove essentially any interference in the said mechanism, in (B) a nutrient medium favouring the multiplication of the blastospores until a population greater than or equal to 104 yeasts/ml is obtained; (2) culturing the yeast blastospores thus obtained in a nutrient medium favouring the filamentation; (3) bringing the filamentous forms thus obtained into contact with a substance (F) chosen from the set consisting of fibrinogen, fibrin, FDPs and mixtures thereof; then (4) assessing the adhesion or non-adhesion of the said substance (F) onto the said filamentous forms. This process is useful for distinguishing pathogenic yeasts from non-pathogenic yeasts.
