PURPOSE:To obtain the subject culture base by putting a vessel filled with a medium into a retort and successively repeating the evacuation and the intro duction of steam while keeping the steam pressure in the second and following steps to a level slightly higher than the saturated steam pressure corresponding to the temperature of the medium, thereby saving the power, labor and time for the sterilization of the base. CONSTITUTION:Culture bags 4 (e.g. polypropylene bag) filled with a medium raw material 1 are stacked on trucks 9, 9' and put into an oven 5. A valve 14 of a vacuum-suction pipe 13 is opened to evacuate the oven 5. The pipe 1e is closed, a valve 11 of a steam pipe 10 is opened to fill the oven 5 with steam of e. g. 1.5-2.7kg/cm2 pressure and the steam is maintained at 110-130 deg.C for several minutes. The steam pipe 10 is closed and a vacuum pump 7 is ener gized to evacuate the oven 5 to a level higher than the saturated steam pressure coresponding to the temperature of the medium and lower than the pressure 1.3 times the saturated pressure. The suction of air and the introduction of steam are repeated several times to obtain the objective culture base.
