PURPOSE:To readily prepare the subject DNA by screening as cDNA library originated from cells producing alpha-fetoprotein (alpha-PH) with a probe comprising a DNA in the enhancer region of alpha-PH gene. CONSTITUTION:An alpha-PH-producing cell such as human liver cell HuH-7 is cultured, and the produced cells are extracted to prepare (a) a mRNA. The component (a) is fractionally extracted to prepare (b) a poly(A) RNA. The component (b) is converted into a double-strand cDNA and introduced into a phage to prepare (c) a recombined phage. A cDNA library obtained by multiplying the component (c) is screened to prepare a DNA sequence which has an amino acid sequence of the formula, etc., or an amino acid sequence of the formula in which the No.508 base is changed into some other amino acid, and which codes a protein specifically combined with the enhancer of alpha-PH gene.
