PURPOSE:To separate a microorganism producing a capsular polysaccharide readily with ease, by cultivating kefir grains in a separating culture medium consisting essentially of wine or wine and polyoxyethylene sorbitan monooleate. CONSTITUTION:Kefir grains are anaerobically cultivated in a culture medium containing wine or wine and polyoxyethylene sorbitan monooleate as essential components and further milk whey, glucose, galactose, agar, etc., to separate the aimed microorganism producing a capsular polysaccharide, e.g. WT-1 of the genus Lactobacillus (FERM-P 8187) from the above-mentioned culture medi um.0.
