PURPOSE: To mass-produce rice plant seedlings from a rice plant callus in which the rice plant body can efficiently be regenerated with a similar efficiency as that of gyratory culture without requiring a field in growing seeds by aerating the rice plant callus with a gas obtained by adding oxygen to air and carrying out the liquid culture. ;CONSTITUTION: A full ripe rice plant seed is peeled and then treated with a 70% ethanol for 5sec, further treated with sodium hypochlorite for 30min, sterilized, sufficiently washed with sterile water, bedded on a callus induction culture medium and cultured at 27°C in the dark for 8 days to extract a callus produced from a part of a blastodisk. The extracted callus is then transplanted to the same culture medium as the callus induction culture medium to further continue the culture for 14 days. The cultured callus is transplanted to a liquid proliferation culture medium and aerated with a gas which is obtained by adding oxygen to air to provide 40-100% oxygen content. Thereby, the liquid culture is carried out and the rice plant body can be regenerated with a similar efficiency as that of gyratory culture with a flask.
