Process for the preparation and storage of biological samples prior to the measurement of concentrations of Mg2 + ionized in order to obtain accurate and reproducible values. Also disclosed are a new accurate measurement method of magnesium ion concentrations in biological samples, a concentration control method of Mg2 + ionized and reports Ca2 +: Mg2 + ionized in a patient, method useful in the diagnosis and prognosis of disease states including cardiopulmonary bypass surgery, hypertension, abnormalities occurring during pregnancy, head trauma and fetal anomalies, and a compositioin and a method of treating a patient with Mg2 + ionized or Mg 2+ and Ca 2+ ionized.
