PURPOSE:To obtain Escherichia coli useful for industrial production of human preformed chemical mediator, having excellent manifestation efficiency of protein production, by transforming Escherichia coli with a promoter having a specific base sequence. CONSTITUTION:DNA fragment of a natural promoter such as a promoter of Escherichia coli is combined to give an improved promoter which has -35 range in a base sequence shown by formula I (X and Y are any base pair of TA, GC, AT or CG) and/or -10 range in a promoter of a base sequence shown by formula II, further a range between the -35 range and the -10 range consisting of 15-17 base pairs and =60% base pairs between the range, comprising GC salt pairs. Then a structural gene such as superoxide dismutase is bonded to the downstream of any one promoter of the promoters having the base sequences shown by the formula III-formula V to give a vector. Then the vector is introduced to plasmid pBR322 and Escherichia coli is transformed with the prepared plasmid to give Escherichia coli such as Escherichia coli K12 strain.
