Process for identifying and/or cloning functional transcriptional promoters in a host comprises: (a) preparing a DNA library of a starting cell; (b) inserting a transposon bearing a selectable marker into the DNA; (c) selecting clones expressing the marker; and (d) isolating DNA fragments comprising the promoter regions of the selected clones. Also claimed are a process for producing a recombinant protein by expression of a DNA sequence coding for the protein under the control of a promoter obtained by the above process and a Kluyveromyces DNA library into which a MiniMu transposon contg. a selectable marker has been inserted. USE/ADVANTAGE - The promoters may be used for expression of genes coding for pharmaceutical or food proteins or genes for the biosynthesis of metabolites, e.g. amino acids, vitamins or antibiotics. The process can be applied to all types of starting cells without prior knowledge of the function or structure of the gene naturally expressed by the promoter.
