QUANTITATIVE DETERMINATION METHOD BY ENZYME IMMUNOREACTION OF CORTISOL IN SALIVA

PURPOSE: To achieve an accurate determination of cortisol in saliva with high sensitivity and high reproducibility without use of any radioactive isotope by mixing the saliva with an enzymejmarked cortisol and an anti-cortisol antibody to react. ;CONSTITUTION: Saliva is mixed with an enzyme-marked cortisol and an anti- cortisol antibody to react. Then, a mixture of a first reaction product of eortisol contained in the saliva and the anti-cortisol antibody contained in the saliva and a second reaction product of the enzyme-marked cortisol and the anti- cortisol antibody is separated from the anti-cortisol antibody left unreacted. Then, a color former and a coloring assistant are added to the mixture to measure absorbance. The enzyme used acts as a catalyst for enzyme reaction between the color former and the coloring assistant and includes peroxidase, alkali phosphatase and the like. The anti-cortisol antibody is produced in vivo by injecting cortisol (antigen) into blood of an immune animal such as rabbit, goat or the like and taken out in the form of serum.;COPYRIGHT: (C)1995,JPO
