The invention relates to a graduated selectivity medium for the preferential growth of Salmonella present in conditions of diminished vitality or in limited number in biological samples or miscellaneous foodstuffs, to be used as a preliminary stage prior to the use of specific isolation and identification systems. The invention further relates to a container provided with a screw cover on whose lower surface is fixed a blade which enters the container. The screw cover can be replaced at a second stage with another similar screw cover which has attached to it a test tube, also closed with a cap, preferably a screw cap, which has a porous septum in its base and a lateral opening close to the cap, instead of a blade. After the material in question has been suspended within the container, the container cover with the blade is removed and replaced by the cover with the test tube. After suitable incubation the screw cap of the test tube is opened and sampling is performed for subsequent transplanting to a selective medium directly from the test tube without further operation involving the faeces.
