PURPOSE:To provide the title method for determining substances as markers for morbid states primarily in diagnosing diseases, etc., using NADH kinase high in specificity to NADH. CONSTITUTION:Using NADH kinase specific to NADH, NADH alone is phosphorylated in a mixture system containing both NAD+ and NADH into NADPH, which is then guided to cycling reaction, thus determining a minimal amount of NADH in high sensitivity. Similarly, a minimal amount of XTP can also be determined in high sensitivity. The present determination can be made without exerting influence of NAD+, rendering itself useful for diagnosing diseases, etc.
