PURPOSE:To obtain horseradish peroxidase useful as a clinical examination agent and an agent for enzyme immunoassay, by preparing a plasmid containing the titled structural gene from a cultured cell of horseradish, transforming a cell with said plasmid and culturing the obtained host cell. CONSTITUTION:Whole RNA is extracted from cultured cell of horseradish (preferably tissue cultured cell of root) by conventional method and the obtained crude RNA is adsorbed to an activated oligo(dT)cellulose column and eluted to obtain poly(A)+RNA, from which a cDNA library of cultured horseradish cell is produced. A strain transformed with a plasmid containing the structural gene of peroxidase is selected from the cDNA library using a probe consisting of a DNA synthesized based on the amino acid sequence of horseradish peroxidase, and the base sequence of the obtained strain is determined. A DNA containing said structural gene is produced and linked to a proper manifestation vector to obtain a recombinant plasmid.
