PROBLEM TO BE SOLVED: To identify a functional ligand to a receptor, by isolating an estimated ligand-coupled domain DNA sequence, replacing the sequence with a known ligand responsive receptor DNA coupled region, introducing a cell deficient in a receptor and expressing. SOLUTION: An estimated ligand-coupled domain and a DNA sequence containing the domain are isolated. The DNA coupled domain region of the DNA sequence is replaced with a DNA coupled domain region from a known ligand responsive receptor protein to constitute a chimera gene. A proper host cell deficient in a receptor is transfected with the chimera gene and a reporter gene coupled with a hormone responsive element having actions of being activated in the DNA coupled domain region of a receptor protein to be encoded by the chimera gene. The host cell is detected by a candidate ligand to be bound with the ligand coupled domain region of a chimera receptor protein and a ligand capable of inducing the protection of a reporter gene product is selected to identify a ligand to a receptor.
