PROBLEM TO BE SOLVED: To replicate nucleic acid with significantly improved replication efficiency of strand-displacement replication reaction by incorporating a strand- displacement DNA polymerase and a single strand DNA-binding protein into the system in an amount sufficient to increase the replication efficiency of a long target. SOLUTION: In this replication of nucleic acid, a long target is replicated by the strand-displacement replication reaction with increased replication efficiency. In the strand displacement replication reaction, a strand-displacement DNA polymerase and a single strand DNA-binding protein as gp32, rpa-1, rim-1,, gp2.5 and rp5 and the like participate and this single strand DNA-binding protein is used in an amount sufficient to increase the replication efficiency in comparison with that of the long target under the conditions that there is no this single strand DNA-binding protein to carry out the strand displacement amplification(SDA) thereby replicating nucleic acid with significant improvement of the amplification efficiency of the long target that cannot be efficiently amplified.
