Diagnosis of coeliac disease - using gliadin fragment to induce endomysial antibody formation in organ culture

A method for in vitro diagnosis of coeliac disease comprises: (a) taking biopsy samples of enteral mucosa from the distal duodenum by gastroscopy; (b) immersing a first sample in an organ culture medium as a control; (c) immersing a second sample in an organ culture medium containing either a peptic-tryptic digest of gliadin or a gliadin fragment containing the amino acid sequence Gln Gln Gln Pro; (d) incubating the cultures at 37[deg]C under a sterile atmosphere of 95% oxygen and 5% CO2 for 24-72 hours; and (e) subjecting the culture supernatants to an immunofluorescence assay for endomysial antibodies.
