PROBLEM TO BE SOLVED: To provide a method enabling classification (allele typing), at genetic level, of A-antigen subtypes whose discriminative classification has been impossible by conventional means, through resolving problems on assaying HLA-A locus antigen typing by conventional serological method. and to provide a kit or reagent therefor. ;SOLUTION: This method is a combination of A'-method with B-method. The A'-method is as follows: using a pair of primers specific to the base sequence of at least one specific HLA-A allele, and also using a set of pairs of primers corresponding to the respective groups each specific to a base sequence common to respective genes in at least one specific group consisting of a specific HLA-A allele group, PCR is performed, and a product afforded by selective amplification is developed by electrophoresis and detected; and the B-method is as follows: the amplified product of the HLA-A allele group in each specific group is treated with a restriction enzyme affording specific breakage site(s) or a restriction enzyme affording no breakage site at all, and the resulting product is then developed by electrophoresis and detected.;COPYRIGHT: (C)1999,JPO
