PURPOSE:To obtain a differentiation and proliferation promoting glycoprotein for monocyte and macrophage by culturing human myelocytic leukemia cell under protein-free or serum-free condition, concentrating the supernatant liquid of the culture product, pretreating at pH2.5-5.0 and separating insoluble fraction. CONSTITUTION:A human myelocytic leukemia cell is cultured under protein-free or serum-free condition, the supernatant liquid of the culture product is concentrated, pretreated at pH2.5-5.0 and the insoluble fraction is separated. The treated liquid is made to contact with an anion exchange material at pH5.0-7.5 to effect the adsorption of valuable substances to the exchange material and a fraction eluted by a 0.05-0.3M inorganic salt solution is collected. The fraction is concentrated and made to contact with a gel-filtration agent to collect a fraction having a relative eluted liquid volume of 1.5-1.8. The eluted liquid is subjected to reversed phase ion-exchange chromatography to obtain the objective marrow cell differentiation and proliferation factor.
