A method of interrupting the expression of amacromolecular synthesis operon in bacteria comprising thestep of binding an antisense oligonucleotide to a singlestranded DNA or to a mRNA transcribed from themacromolecular synthesis operon. The antisenseoligonucleotide can be either sequence specific to a uniqueintergenic sequence or a sequence specific to a bacterialhomologous sequence. By interrupting the expression of themacromolecular synthesis operon bacterial infections can betreated. Specific antisense oligonucleotides aredisclosed. The ability of the antisense oligonucleotide tobind the mRNA or single stranded DNA also allows theidentification of the bacteria by using a unique intergenicantisense oligonucleotide to bind to the single strandedDNA or to the mRNA transcribed from the macromolecularsynthesis operon. A method for competitively inhibitingthe protein products of the MMS operon witholigonucleotides is also disclosed. Methods of identifyingunique intergenic sequences are also disclosed.
