摘要
The present invention is a method of developing a virus resistant plant by introducing a nucleotide sequence of domain 1 present in a replication enzyme of tobacco mosaic virus, and a plant having a novel trait of virus resistance developed by the method.;The domain 1 sequence was cut out from the complementary DNA (cDNA) containing the entire gene of the tobacco mosaic virus, inserted into the plant expression vector to construct a recombinant plasmid, and then the domain 1 sequence was introduced into the plant by co-culture with Agrobacterium. .;Genetic analysis confirmed the introduction and expression of the domain 1 sequence in the transgenic plants. As a result of investigating the degree of virus resistance in the transgenic plants, all transgenic plants introduced with the domain 1 sequence were completely resistant to the virus, and at very high virus inoculation concentration (1000 µg / ml) and tobacco mosaic virus Resistance was maintained even when inoculated with RNA (TMV RNA, 10 µg / ml).;The presence of domain 1 sequences in transformed tobacco plants completely inhibits the systemic disease development and propagation of the virus associated with tobacco mosaic virus infection.;Although the present invention has been identified only for tobacco mosaic virus, the domains of plant RNAs, such as cucumber mosaic virus, alfalfa mosaic virus, potex virus, bromovirus, forty virus, lateovirus, etc., which are not flexible to each other, have been identified. Since there is a motif similar to 1, a strategy using the domain 1 sequence may be applied to the breeding of plants resistant to these viruses.
