PURPOSE: DNA sequence specific to anther gene expression is provided which can be used to develop male sterile plant for the production of hybrid seed. CONSTITUTION: cDNA library of rice(Oryza sativa L.) anther is hybridized with leaf cDNA library and anther specific cDNA clone is obtained. The expression of gene joined with this cDNA is initiated at tatrad stage and reached maximum at vacuolated pollen stage. Therefore, gene expression at tapetum of anther, endothecium, or connective tissues is induced by this cDNA sequence. The coding sequences joined to anther specific promoter used to inhibit development of pollen are RNase, DTA, TURF-13, petite lyase, gin recombines, iaaL or cytA toxin. Promoter sequence is ligated with additional sequence including part of exon or intron. Another cDNA clone which shows 80% of sequence similarity with previously described cDNA sequence is also isolated. The transgenic plant produced with these cDNA clone is male sterile and can be applied to rice, wheat, sweet corn and sorghum.
