PHARMACEUTICAL COMPOSITION TREATING FOR COLLAGEN-INDUCED ARTHRITIS CONTAINING TYPE II COLLEGEN PEPTIDE

PURPOSE: A medicine for collagen arthritis containing peptide of type II collagen(CII) or its analog as an effective ingredient is provided, which inhibits mammal collagen-induced arthritis(CIA) effectively by inducing oral immune tolerance. CONSTITUTION: A process for the preparation of II-type collagen peptide by Trentham is comprised of: collecting cartilage from the cow thighbone, washing with chilled distilled water, homogenizing with the Polytron homogenizer, suspending in 1M NaCl/50mM Tris-HCl(pH 7.6), and shaking for 3days at 4deg.C; centrifuging at 10,000xg, 4deg.C, and gathering precipitates as non-soluble CII; suspending the non-soluble CII in a solution of acetic acid and pepsin, shaking for 3days at 4deg.C, and adding NaOH to inactivate the pepsin; centrifuging the gelled soluble CII, and gathering the precipitates; and dissolving the precipitates in acetic acid, centrifuging to get upper layer, processing dialysis in deionized water, freeze-drying and storing at 4deg.C.
