Transgenic mice containing Human Ig lambda locus

Transgenic mice were created carrying a 380kb region of the human immunoglobulin . g. light (L) chain locus, in germline configuration, using a yeast artificial chromosome (YAC) which comprised at least 15 variable (V) region genes all J. g. -C. g. segments and the 3' downstream enhancer. The resulting mice had ratios of . k. :. g. chains resembling that of humans. Mouse embryonic stem cells containing the HuIg YAC were injected into mouse blastocysts to create chimeric mice expressing the transgene.
