The present invention is directed to vaccine compositions containing pathogenic bacteria, such as Salmonella, having non-reverse genetic mutations that alter DNA adenine methylase (Dam) activity, and methods using these compositions to elicit an immune response. The invention also provides methods for preparing vaccines, as well as screening methods to identify agents that may have antibacterial activity.
