An antibody or antigen-binding fragment produced by a hydridoma cell line deposited as ATCC accession number HB-12195 having a binding specificity for a naturally occurring mammalian C-C Chemokine Receptor 3 protein (CKR-3 or CCR-3) that inhibits the binding of a ligand to the mammalian CKR-3 site. Nucleic acid constructs for the production of recombinant CKR-3 polypeptides and antibodies reactive with the receptors, which are useful in research and diagnostic applications. Also provided are methods of use of the nucleic acids, proteins, and host cells to identify ligands, inhibitors (e.g., antagonists) or promoters (agonists) of receptor function. The administration of the antibody or antigen-binding fragment inhibits or promotes receptor function to an individual in need of therapy for selective modulation of leukocyte function, which is useful in a variety of inflammatory and autoimmune diseases, or in the treatment of infections.
