PURPOSE: Disclosed is a concentration method of micromolecules with macromolecules separation using an on-line reciprocating size exclusion chromatography. CONSTITUTION: The equipment system is composed of a tank (10), a pump (31,32), a column (40) of size exclusion chromatography, a valve (21,22), a solvent tank (51), a tank (52) for macromolecule, a tank (53) for micromolecule, and a timer. Using the chromatography, the operation steps are carried out continuously by the following procedures: an induction step of the mixture solution into the column (40) using the pump (31), a solvent recovery step into the solvent tank (51), a macromolecule recovery step, a reverse flow control step of the solution into the column (40) using a pump (32), and a mixture solution recovery step into the tank (10). The packing material for the column (40) is one of Sephadex G series, BioGel P series, Sephadex LH series, Sepharose series, or Sephacryl series.
