Parallel sequencing of at least two different nucleic acids (I) and (II) in a mixture. Parallel sequencing of at least two different nucleic acids (I) and (II) in a mixture, involves a surface being prepared with islands of nucleic acids (designated tertiary nucleic acid, tNA) of the same sort, then the opposite strands (tNA') are produced, and these are extended by one nucleotide (nt) that is protected at the 2'- or 3'-position, to prevent further extension, and carries an identifiable label. The incorporated nt is identified, then the protecting group removed and the label either removed or altered. The extension step, and all subsequent steps, are repeated as many times as needed to provide the required sequence information.
