Rapid detection of DNA single-base substitution (SBS) using peptide nucleic acid (PNA) probes is realized in a droplet-handling microfluidic device. The device can generate 420-pL droplets of sample solution with high accuracy (±2 pL) and conduct droplet-based reaction and following electrophoresis process for the analysis of the reaction products. All the liquid handlings were implemented by pneumatic manipulation through microcapillary vents on the device. As a demonstration, it is shown that R553X mutation on Cystic Fibrosis Transmembrane Regulator (CFTR) gene can be detected within 1.5 min.
