PURPOSE:To obtain an inexpensive and chemically stable column filler by treating alkyl group-coupl silica gel in an aqueous solution of alkylsulfuric acid alkali metal salt and copper sulfate. CONSTITUTION:The title column filler is obtained by subjecting modified silica gel to solid-liquid separation and washing with water after making an arbitrary alkyl group-coupl silica gel sold on the market as a filler for high precision liquid chromatography (LPLC) to react in an aqueous solution of alkylsulfuric acid alkali metal salt expressed by R1SO2Na (where, R1 represents the alkyl group), such as sodium or potassium salt and copper sulfate. When a column packed with this filler is used, amino acid which is high in polarity and low in UV absorption sensitivity can be directly separated in a stable acidic aqueous solution by prolonging the retention time without changing in advance the amino acid to a derivative of phenyl isothiocyanate, etc. Therefore, highly accurate analysis can be carried out.
