PURPOSE: A DNA cassette for producing a secretory recombinant trimer of TRAIL protein, tetracycline/doxycycline-inducing adeno-related virus vector, a combination of them and the gene therapy are provided, thereby mass-producing the secretory recombinant trimer of TRAIL protein. CONSTITUTION: The DNA cassette for producing a secretory recombinant trimer of TRAIL(tumor necrosis factor-related apoptosis-inducing ligand) protein consists of secretory signal sequence(SS), trimer forming domain(TFD) and TRAIL(114-281) encoding cDNA, in which the secretory signal sequence is SEC2 or SEC(CV); and the trimer forming domain is ILZ. The recombinant expression vector pCMVdw capable of secreting the secretory recombinant trimer of TRAIL protein contains the DNA cassette, SEC2 or SEC(CV) and ILZ. The recombinant expression vector pAAVdw capable of tetracycline/doxycycline-inducible expressing the foreign protein contains SEC2 or SEC(CV) and ILZ. The transformed cell line is produced by using the vector pCMVdw and pAAVdw.
