摘要

PURPOSE: A method of cryopreserving an unfertilized oocyte by quick and simple operating processes without sophisticated or expensive laboratory equipment and trained technique is provided. By the use of the frozen unfertilized oocyte using the cryopreserving method, an elite cow having a dominant gene is produced. CONSTITUTION: The cryopreservation method comprises: treating an unfertilized oocyte in a PBS solution containing ethylene glycol and FBS for 4 to 6min; treating the treated unfertilized oocyte in a vitrification solution containing FBS, ethylene glycol and sucrose for 25 to 35sec; dropping the treated unfertilized oocyte into a straw; and treating the straw with liquid nitrogen. The unfertilized oocyte frozen according the vitrification cryopreservation method is thawed, fertilized in vitro with dominant sperm having a desired dominant gene and then cultured. Thereafter, the cultured fertilized oocyte is transferred to the recipient finished with a synchronization of estrus and induced to delivery after identification of pregnancy of the recipient.
