The invention relates to the translation of RNAs transcribed in the nematode worm IC. elegans/I using a bacteriophage polymerase such as T7 polymerase. This is achieved by the introduction of a trans-splice recognition site recognised by an SL1 trans-splice recognition sequence into the DNA template transcribed by the bacteriophage polymerase i.e a bacteriophage promoter is operably linked to an outron sequence. The outron sequence may be isolated from natural IC. elegans/I genes or a synthetic outron sequence may be synthesized. The bacteriophage promoter and outron sequence can be used in expression vectors and replicable cloning vectors.
