"IMMUNOGENIC COMPOSITION ADAPTED FOR ADMINISTRATION TO A HUMAN HOST MANAGER FOR THE TREATMENT OR PREVENTION OF ATHEROSCLEROSIS, VACCINE, AND METHOD OF TREATMENT OR PROPHYLAXIS OF ATHEROSCLEROSIS". The invention relates to new vaccinotherapies and prophylactic treatments of atherosclerotic diseases. Thus, immunogens are provided comprising apolipoproteins, or fragments or derivatives thereof, which, in its native form, apolipoprotein has at least one of the following activities: (a) inhibiting the binding of Apolipoprotein B to its receptor, and / or ( b) inhibition of lipoprotein lipase. A preferred example of such an apolipoprotein is Apolipoprotein C-III (ApoCIII). The vaccines of the present invention, comprising said immunogens, are potent in preventing, or reducing, the formation of atherosclerotic plaques over prolonged periods of time, thereby reducing the potential for atherosclerosis leading to coronary or cerebrovascular disease. Thus, a novel method of treating atherosclerosis by selective inhibition of specific apolipoprotein activity in a human host is provided, and in a preferred aspect of the invention, apolipoprotein is ApoCII, comprising administering an agent that results in selective inhibition of activity. apolipoprotein to a human. Methods of treating or preventing atherosclerosis by active vaccination or passive vaccination by administering to a patient an antibody capable of binding to an apolipoprotein which has at least one of the following activities are also provided: inhibiting the binding of Apolipoprotein B to its receptor and / or (b) inhibiting lipoprotein lipase; and, in this aspect of the invention, the antibody preferably binds and abrogates ApoCIII activity. Also provided is the use of the immunogens or vaccines of the present invention in medicine, and methods of their production.
