Improved methods for amplifying nucleic acids can reduce non-specificamplification and minimize the effects of contam-ination of nucleic acid amplification reaction assays due to amplified productfrom previous amplifications. The methods involvethe introduction of unconventional nucleotide bases into the amplificationreaction products and treating the products by enzy-matic (e.g., glycosylases) and/or physical-chemical means to render theproduct incapable of acting as a template for subsequentamplifications.
